چکیده

Background The human genome codes for 49 members of the large ABC protein family. Most of them are membrane transporters. The first structures of exporters have been solved in recent years. Although a remarkable achievement, some uncertainty remained with respect to the physiological conformation of the transporter, which seems not to be fully compatible with all biochemical evidence. During crystallization, the transporter is extracted from its native membrane environment and solubilized by surfactants. This procedure might have influenced the transporters’ conformation and lead to the structures observed in the crystals. We focus on the multidrug resistance transporter P-glycoprotein, which prevents xenotoxic compounds from entering the body and from penetrating into the brain, and confers resistance to chemotherapy.

Bcl-2 changes conformation to inhibit Bax oligomerization.

Bcl-2 inhibits apoptosis by regulating the release of cytochrome c and other proteins from mitochondria. Oligomerization of Bax promotes cell death by permeabilizing the outer mitochondrial membrane. In transfected cells and isolated mitochondria, Bcl-2, but not the inactive point mutants Bcl-2-G145A and Bcl-2-V159D, undergoes a conformation change in the mitochondrial membrane in response to a...

A structural model for the open conformation of the mdr1 P-glycoprotein based on the MsbA crystal structure.

The validity of the structure of the Escherichia coli MsbA lipid transporter as a model from the mdr1 P-glycoprotein has been evaluated. Comparative sequence analyses, motif search and secondary structure prediction indicated that each of the two P-glycoprotein halves is structurally similar to the MsbA monomer and also suggested that the open dimer structure is valid for P-glycoprotein. Homolo...

Inhibitory effect of clemastine on P-glycoprotein expression and function: an in vitro and in situ study

Objective(s):Transporters have an important role in pharmacokinetics of drugs. Inhibition or induction of drug transporters activity can affect drug absorption, safety, and efficacy. P-glycoprotein (P-gp) is the most important membrane transporter that is responsible for active efflux of drugs. It is important to understand which drugs are substrates, inhibitors, or inducers of P-gp to minimize...

P-24: Beta Defensin 126 and Its Role in Intra-Uterine Insemination Outcome

Background: Human β-defensin 126 is a small cationic glycoprotein that coats the plasma membrane of sperm during epididymal transit. It provides protection for sperms from infection-causing microbes and against the female immune system. Defensin remains on the sperm until sperm become capacitated in the female reproductive tract. DEFB126 gene is located on the subtelomeric end of 20p13 in human...

Unfolding events in the water-soluble monomeric Cry1Ab toxin during transition to oligomeric pre-pore and membrane-inserted pore channel.

The insecticidal crystal (Cry) proteins produced by Bacillus thuringiensis undergo several conformational changes from crystal inclusion protoxins to membrane-inserted channels in the midgut epithelial cells of the target insect. Here we analyzed the stability of the different forms of Cry1Ab toxin, monomeric toxin, pre-pore complex, and membrane-inserted channel, after urea and thermal denatur...
